This study examined the influence of high glucose on oxidative stress-induced hepatic cell death. Hc cells, a normal human hepatocyte-derived cell line, were cultured in normal-to-high glucose (5.5-22 mM)-containing medium with varying concentrations (0.01-1 mM) of hydrogen peroxide. In certain experiments, cyclosporine A (CyA), which inhibits the mitochondrial permeability transition (MPT) pore, or Z-VAD-FMK (z-VAD), a pan-caspase inhibitor, were added to the medium. Cell viability was evaluated using a colorimetric assay. The mode of cell death was determined by nuclear staining methods using Hoechst 33258 and Sytox green. Neither high glucose (22 mM) nor 0.05-0.5 mM of hydrogen peroxide alone killed Hc cells. However, a combination of the two induced cell death, causing the nuclei of Hc cells to become expanded rather than condensed, and the nuclear membrane to become weak. CyA, but not z-VAD, blocked cell death. These results suggest that a high glucose condition may cause human hepatocytes to undergo hydrogen peroxide-induced necrotic cell death.", "subitem_description_type": "Abstract"}]}, "item_2_description_63": {"attribute_name": "引用", "attribute_value_mlt": [{"subitem_description": "Molecular Medicine Reports, 1(3), pp.379-385; 2008", "subitem_description_type": "Other"}]}, "item_2_publisher_33": {"attribute_name": "出版者", "attribute_value_mlt": [{"subitem_publisher": "Spandidos Publications"}]}, "item_2_source_id_7": {"attribute_name": "ISSN", "attribute_value_mlt": [{"subitem_source_identifier": "17912997", "subitem_source_identifier_type": "ISSN"}]}, "item_2_source_id_8": {"attribute_name": "EISSN", "attribute_value_mlt": [{"subitem_source_identifier": "1791-3004", "subitem_source_identifier_type": "ISSN"}]}, "item_2_version_type_16": {"attribute_name": "著者版フラグ", "attribute_value_mlt": [{"subitem_version_resource": "http://purl.org/coar/version/c_970fb48d4fbd8a85", "subitem_version_type": "VoR"}]}, "item_creator": {"attribute_name": "著者", "attribute_type": "creator", "attribute_value_mlt": [{"creatorNames": [{"creatorName": "Shibata, Hidetaka"}], "nameIdentifiers": [{"nameIdentifier": "62522", "nameIdentifierScheme": "WEKO"}]}, {"creatorNames": [{"creatorName": "Ichikawa, Tatsuki"}], "nameIdentifiers": [{"nameIdentifier": "62523", "nameIdentifierScheme": "WEKO"}]}, {"creatorNames": [{"creatorName": "Nakao, Kazuhiko"}], "nameIdentifiers": [{"nameIdentifier": "62524", "nameIdentifierScheme": "WEKO"}]}, {"creatorNames": [{"creatorName": "Miyaaki, Hisamitsu"}], "nameIdentifiers": [{"nameIdentifier": "62525", "nameIdentifierScheme": "WEKO"}]}, {"creatorNames": [{"creatorName": "Takeshita, Shigeyuki"}], "nameIdentifiers": [{"nameIdentifier": "62526", "nameIdentifierScheme": "WEKO"}]}, {"creatorNames": [{"creatorName": "Akiyama, Motohisa"}], "nameIdentifiers": [{"nameIdentifier": "62527", "nameIdentifierScheme": "WEKO"}]}, {"creatorNames": [{"creatorName": "Fujimoto, Masumi"}], "nameIdentifiers": [{"nameIdentifier": "62528", "nameIdentifierScheme": "WEKO"}]}, {"creatorNames": [{"creatorName": "Miuma, Satoshi"}], "nameIdentifiers": [{"nameIdentifier": "62529", "nameIdentifierScheme": "WEKO"}]}, {"creatorNames": [{"creatorName": "Kanda, Shougo"}], "nameIdentifiers": [{"nameIdentifier": "62530", "nameIdentifierScheme": "WEKO"}]}, {"creatorNames": [{"creatorName": "Yamasaki, Hironori"}], "nameIdentifiers": [{"nameIdentifier": "62531", "nameIdentifierScheme": "WEKO"}]}, {"creatorNames": [{"creatorName": "Eguchi, Katsumi"}], "nameIdentifiers": [{"nameIdentifier": "62532", "nameIdentifierScheme": "WEKO"}]}]}, "item_files": {"attribute_name": "ファイル情報", "attribute_type": "file", "attribute_value_mlt": [{"accessrole": "open_date", "date": [{"dateType": "Available", "dateValue": "2020-12-23"}], "displaytype": "detail", "download_preview_message": "", "file_order": 0, "filename": "MMR_1_3_379.pdf", "filesize": [{"value": "202.8 kB"}], "format": "application/pdf", "future_date_message": "", "is_thumbnail": false, "licensetype": "license_free", "mimetype": "application/pdf", "size": 202800.0, "url": {"label": "MMR_1_3_379.pdf", "url": "https://nagasaki-u.repo.nii.ac.jp/record/16461/files/MMR_1_3_379.pdf"}, "version_id": "72ffbe48-6036-48a2-9502-0a11f1f0eebe"}]}, "item_keyword": {"attribute_name": "キーワード", "attribute_value_mlt": [{"subitem_subject": "high glucose", "subitem_subject_scheme": "Other"}, {"subitem_subject": "reactive oxygen species", "subitem_subject_scheme": "Other"}, {"subitem_subject": "hepatocyte cell death", "subitem_subject_scheme": "Other"}, {"subitem_subject": "necrosis", "subitem_subject_scheme": "Other"}, {"subitem_subject": "cyclosporine A", "subitem_subject_scheme": "Other"}]}, "item_language": {"attribute_name": "言語", "attribute_value_mlt": [{"subitem_language": "eng"}]}, "item_resource_type": {"attribute_name": "資源タイプ", "attribute_value_mlt": [{"resourcetype": "journal article", "resourceuri": "http://purl.org/coar/resource_type/c_6501"}]}, "item_title": "A high glucose condition sensitizes human hepatocytes to hydrogen peroxide-induced cell death", "item_titles": {"attribute_name": "タイトル", "attribute_value_mlt": [{"subitem_title": "A high glucose condition sensitizes human hepatocytes to hydrogen peroxide-induced cell death"}]}, "item_type_id": "2", "owner": "2", "path": ["9"], "permalink_uri": "http://hdl.handle.net/10069/22341", "pubdate": {"attribute_name": "公開日", "attribute_value": "2009-11-09"}, "publish_date": "2009-11-09", "publish_status": "0", "recid": "16461", "relation": {}, "relation_version_is_last": true, "title": ["A high glucose condition sensitizes human hepatocytes to hydrogen peroxide-induced cell death"], "weko_shared_id": -1}
